I am curious as to all your thoughts on how to get the best results when repainting the numbers on an etched 12" by 27" 10 gallon cylinder. What method do you feel produces the best results?

First off you are going to want to make sure that all the old paint is gone.
Once that is accomplished, I would suggest masking around the numbers. You don't have to mask right up to the numbere - you can just make a square around them with a small bit of the smooth glass still showing.
Mask the rest of the cylinder from overspray and shoot the 'squares' with the color you want using a spray can.
After the paint completely drys, unmask the entire cylinder and use a straght-edge razor blade to scrape any paint off the smooth portion of the glass leaving the paint in the numbers.
